    Effect of Kunshen Granule on the Expression of VEGF-C、VEGFR-3 of Human Gastric Carcinoma in Nude Mice Model

    Objective To probe the effect of Kunshen Granule on the expression of vascular endothelial growth factor-C(VFGF-C) and vascular endothelial growth factor receptor-3(VFGFR-3). Methods 40 BALB/C nude mice were randomly divided into 4 groups: negative control group, 5-Fu group, Kunshen large dose group and small dose group. The nude mice except the negative group were implanted human cancer cell SGC-7901 on stomach near the greater curvature side by means of plastering "OB" gum. And then nude mice were treated with normal saline, 5-Fu, Kunshen large dose and small dose respectively. After six weeks, the animals were anesthetized, sacrificed and the local growth transplanted tumor were observed. The expressions of VEGF-C and VEGFR-3 in the local transplanted tumor tissues were detected and analyzed semi-quantitatively by means of Envision immunohistochemistry staining. Results ①The transplanted tumors in 40 nude mice were growing well, and some of them had enlarged abdominal lymph nodes, lymph nodes metastasis rates of 5-Fu group, Kunshen large dose group and small dose group were 50%,40%,60%, significantly lower than negative control group, which was 100%.②Compared to negative control group, the expressions of VEGF-C and VEGFR-3 in 5-Fu group, Kunshen large dose group and small dose group were significantly decreased, the difference had a statistical significance(P <0.01). Difference of the expression of VEGF-C between the Kunshen large dose group and 5-Fu group had a statistical significance(P <0.05). Difference of the expression of VEGF-C and VEGFR-3 between the Kunshen large dose group and Kunshen small dose group had a statistical significance(P<0.01).③There is linear correlativity between the expression of VEGF-C, VEGFR-3 and the lymph nodes metastasis nearly(r=0.825, P=0.00). Conclusion Kunshen granule can decreased the expression of the VEGF-C and VEGFR-3, which may be one of the mechanisms inhibiting carcinoma lymph nodes metastasis.

    The Effects of Long Cao Particles on the Expression of ET and NO in Hypertensive Rats' Vessels

    Objective To observe the effects of Long Cao Particles on the expression of ET and NO in hypertensive rats' vessels. Methods Divide the spontaneously hypertensive rats(SHR) into such groups as positive, large dose of Long Cao Particles, small dose of Long Cao Particles, negative and wistar. After a month of intragastric administration, test the expression of ET and NO in the vessels by means of immune tissue S-P chemotherapy and hybridization in situ. Results Long Cao Particles can reduce ET and raise the expression of NO in hypertensive rats' vessels. Conclusion By regulating the expression of ET and NO level in vessels, Long Cao Particles can improve the endothelial function and bring high blood pressure down.

    Objective Preliminary to study the action of anti-dysmenorrhea of Xigui. Methods Record the effect of excitatory action which is caused by the spontaneous activity of uterine smooth muscle strips and the oxytocin through BL-410 system and study its effect to writhing response of mice which is caused by acetic acid and oxytocin, evaluate the anti-dysmenorrhea action of Xigui. Results Xigui can significantly reduce the frequency, amplitude, area and duration of uterine smooth muscle systolic wave of rats, meanwhile, it can significantly restrain the excitatory action of uterine smooth muscle which is induced by the oxytocin(P<0.05 or P<0.01). It also can significantly reduce the times of writhing response which is caused by acetic acid and oxytocin(P<0.05 or P<0.01). Conclusion Xigui has the functions of anti-dysmenorrhea.

    Hemodynamic Effects of Intravenous Oxytocin on Different Painless Abortion Patients

    Objective To explore the hemodynamic effects of Intravenous oxytocin on different painless abortion patients. Methods Select 30 cases of the painless abortion patients, which were divided into groupⅠ(age≥40) and groupⅡ(age<40), each 15 cases. Two groups of patients were under anesthesia operation with propofol. Intravenous oxytocin 10U was injected in 30s after uterine suctions and hemodynamic stability. MAP and HR were recorded before and after oxytocin injection. Results Compared to those before, MAP was decreased significantly and HR was increased fast after oxytocin injection in both groups(P<0.05).The decrease of MAP was slower but lasted for a longer period in group Ⅰ than in group Ⅱ (P<0.05). Conclusion Intravenous oxytocin could make hemodynamic fluctuation on different painless abortion patients, longer in over 40 ones than in young ones and easy to induce higher adverse cardiovascular risk.

    Effective Observation on Chronic Hepatitis B with Syndrome of Liver Stagnation and Spleen Deficiency Treated by Modified Xiaoyaosan

    Objective To observe the clinical effection of modified Xiaoyaosan in treating chronic hepatitis B with Syndrome of liver stagnation and spleen deficiency. Methods 60 patients of chronic hepatitis B with syndrome of liver stagnation and spleen deficiency were chosen for clinical observation, and randomly divided into experimental group and control group. After12 weeks' treatment, to observe the patients' improvements such as clinical symptoms, biochemical indicators, immunological targets, pathogen indicators and so on. Results After the treatment of 12 weeks, the total efficiency of experimental group is 83.33%, and 66.67% of the control group. Experimental group was significantly better than the control group(P <0.05). The experimental group is superior to the control group in the improvement in Syndrome of TCM, biochemical, immunological indicators and HBV-DNA negative conversion rate, and there are statistical differences between the two groups(P <0.05). Conclusion Modified xiaoyaosan can effectively improve Syndrome of TCM, regulate cell immunity, restore liver function and enhance anti-viral response rate.

    Objective To discuss the effect of Yi Shen Wu Fa oral liquid on mice's acute toxicity. Methods The classical method of acute toxicity were used to study on mice's caused by Yi Shen Wu Fa oral liquid. Results Caculated by Yi Shen Wu Fa oral liquid, the MLD is 47.5g·kg-1·d-1, which is equal to 280.0 times of 70kg people's daily dried medicinal herb expenses. The main acute toxicity symptoms of mouse are negligent action, diarrhea and hypokinesia. Compared with the control group,The mice's body-weight gainings were slow after 6 day's administration, which gradually normal after 8 day's administration and there were no other obvious abnormalities. The serum concentration of ALT and AST, the liver-body ratio all increased after 2 hours' administration. Conclusion The mice's acute toxicity of Yi shen wu fa oral liquid could induce the liver injury although could not make the LD50 and MTD, and the mice did not appear dead. It demonstrated that the drug gavages certain dose to mice could cause obvious hepa-injury, which in accordance with the clinical ADR report.

    Objective Study the "dosage-time-toxicity" relationship of hepatotoxicity induced by Yi Shen Wu Fa oral liquid in mice. Methods Study on the "time-toxicity"relationship: Ig Yi Shen Wu Fa oral liquid of certain dosage to mice and the changes of the level of ALT, AST, TBI in serum and organ index of liver, spleen and thoracic gland were tested at different time points after administration. Study on the "dosage-toxicity"relationship: Ig Yi Shen Wu Fa oral liquid of different dosages to mice and the corresponding treatment was performed at 2h after administration in accordance with the method of study on"time-toxicity"relationship. Results The study on the "time-toxicity" relationship indicated that the level of ALT, AST, TBI in serum reached to the peak at 2h after once administration of Yi Shen Wu Fa oral liquid of higher dosage and the liver index rised to its topmost at 12h. They all recovered to normal value at 72h after ig. The study on the "dosage-toxicity"relationship indicated that conspicuous hepartotoxical injury can be induced by Yi Shen Wu Fa oral liquid of (4.46~25)mL·kg-1 and the level of ALT, AST, TBI increased significantly with the increase of dosage. Conclusion Once oral administration of Yi ShenWu Fa oral liquid with large dosage may induce acute hepatotoxical injury in mice and show an obvious "dosage-time-toxicity" relationship.

    Pathologic Research on Hepatotoxicity in Mice Caused by Yi Shen Wu Fa Oral Liquid

    Objective To investigate the pathologic injury degree of hepatotoxicity in mice induced by single administration of Yi Shen Wu Fa oral liquid and provide pathohistological basis for its hepatotoxical injury. Methods The pathologic changes of mice in the study on "dose-time-toxicity"relationship of hepatotoxicity caused by Yi Shen Wu Fa oral liquid was observed with light microscope and image analysis after HE stain. Results The study on the "time-toxicity" relationship indicated that the hepatotoxicity occurred at 2h after administration and expressed obviously in the range from 6h to 24h after administration. The study on the "dose-toxicity"relationship showed that pathohistological injury of different degree can be induced by drug of (10.56~25.00) g·kg-1 and acidophilia, hyalinization, vacuolus were the main pathologic changes. Conclusion Once oral administration of Yi Shen Wu Fa oral liquid with certain dosage may induce acute hepatotoxical injury in mice and show an obvious "dose-time-toxicity"relationship. It can cause pathologic changes such as acidophilia, hyalinization, vacuolus to hepatic cells.

    Objective To establish a method for determing 2, 3, 5, 4-tetrahydroxystibene-2-O-β-D-glucosid 、emodin、emodin monomethyl ether in yishen wufa oral liquids. To explore the relationship of toxicity of Yi Shen Wu Fa oral liquids and Radix Polygoni multiflori from the perspective of index constituents, and to provide method for the quality research of yishen wufa oral liquids. Methods HPLC, stationary phase was C18 column(4.6mm×250mm,5μm), mobile phase of 2,3,5,4′-tetrahydroxystibene-2-O-β-D-glucosid was acetonitrile-water solution(25∶75), detection wavelength 320nm, flow rate1.0mL·min-1 and column temperature25℃; mobile phase of emodin and emodin monomethly ether was methanol-0.1% phosphric acid(80:20), detection wavelength 254nm, flow rate1.0mL·min-1 and column temperature25℃. Results There was good linearity in the range of 0.1μg~10μg(r=0.9989) for 2,3,5,4′-tetrahydroxystibene-2-O-β-D-glucosid、in the range of 0.0168~1.68μg(r=0.9984) for emodin and in the range of 0.0208~1.04μg(r=0.9966)emodin monomethly ether; the average recovery of 2,3,5,4′-tetrahydroxystibene-2-O-β-D-glucosid、emodin、emodin monomethyl ether were 99.33%、99.998% and 99.576%, and RSD were 0.68%, 1.11 and 1.29%(n=5). Conclusion This method is simple, with high sensitivity and less interference, high reproducibility and recovery, can be used for quality control of Yi Shen Wu Fa oral liquids. It is initially confirmed that the toxicity of Yi Shen Wu Fa oral liquids may be related with the content of Stilbene glucoside、Emodin and Emodin monomethyl ether in Radix Polygoni multiflori. So, it should control the dose range of Yi Shen Wu Fa oral liquids and avoid adverse events.

    Objective By studying the 33 cases of abrupt group adverse events of conjunctival hyperemia after surgery in a hospital from August 14 to September 1 in 2008, investigated the analysis method for drugs relevance of the abrupt group adverse events cases. Methods The medicines and the medical device products were investigated and analysed, and an inductive analysis was made. Results The probability of the relevance between medicines and the conjunctival hyperemia of the patients after surgery was excluded, and there was significant relevance between the medical device product, medical biodegradable chitosan preventing postsurgical adhesion, and the conjunctival hyperemia. Conclusion The abrupt group adverse events of conjunctival hyperemia was caused by medical biodegradable chitosan preventing postsurgical adhesion.

    Investigate and Analysis of Adverse Drug Reaction of Seven Traditional Chinese Medicine Injections

    Objective To explore the general pattern and characteristics of adverse reaction induced by seven traditional Chinese medicine injections so as to provide some guidance for clinical rational use of drug. Methods A retrospective analysis was conducted on 607 cases of seven traditional Chinese medicine injections' adverse reaction reported in domestic medical journals included in CHKD database from 1994 to 2010. Results Adverse reaction of seven traditional Chinese medicine injections for cardio-cerebrovascular disease often occurred in the people at the age of over 40 years old. The onest of adverse reaction was usually within 30 minutes after injection. Adverse reaction could involve in multiple organs and systems, and clinical manifestations were various, mainly were allergic reaction. Severe allergic reaction could result in anaphylactic shock. Adverse reaction of every drug had themselves general pattern and characteristics in sex and age and type and so on. Conclusion Doctors and pharmacists should know the pattern and characteristics of adverse reaction, and the use of traditional Chinese medicine injection should be under strict monitoring to reduce the occurrence of serious adverse reaction.

    Objective To study clinic features and regularity of anaphylactic shock induced by lopromide injection, to provide reference for rational use of drug. Methods 30 cases ADR of lopromide injection are collected and analyzed statistically from CNKI articles in 1995~2009. Conclusion The duration of anaphylactic shock with lopromide injection is short and death rate is very high, and should be highly emphasized by clinicians.
