    Ion Chromatography Determination of Glucosamine Calcium Citrate Dispersible Tablets

    Objective To establish an assay method of determination for Glucosamine hydrochloride of Glucosamine Calcium Citrate Dispersible Tablets by ion chromatography(IC). Methods The assay was performed on CarboPacTM PA200 3mm×250mm(with CarboPacTM PA200 3mm×50mm protective column); mobile phase:0.2mol/L NaOH,The flow rate was 0.5mL/min; the column temperaturetemprature 30℃. Results The line ranges of Glucosamine hydro-chloride as partate was 0.01~0.2 mg/L(r=0.9995),The average recoveries of Glucosamine hydrochloride were 99.5%. Conclusion The method is proved to be rapid, accurate, and convenient,and provided a reliable way for evaluating the quality of Glucosamine hydrochloride as Glucosamine Calcium Citrate Dispersible Tablets.

    The Effect of Naloxone in the Endurance of the Mice in the Acute and Repeated Anoxia

    Objective In order to determine the dose and time of naloxone(NLX) treatment in the acute and repeated anoxia conditioning, the study was carried out using mice hypoxic models, about the effects of β-endorphin (β-EP), the reverse of respiratory depression and the change of hypoxic endurance time in mice injected with NLX in the hypoxic condition. Methods The mice were divided into 3 groups.The first group was injected with NC, The second group was injected with0.4mg/mL NLX, The third group was injected with0.04mg/mL NLX. Results Oxygen exhausting time of mice was significantly longer under hypoxic condition than under sham condition, but the oxygen exhausting time of mice from the second group under the hypoxic condition was significantly longer than the first group after injection of 0.4mg/mL NLX; there was a significant decrease in the 0.04mg/mL NLX mice compared to those in the first-group mice. Conclusion NLX can block the effect of β-EP released, reverse the respiratory depression, but the NLX in different dose make different effect in the endurance of the mice in the acute and repeated anoxia condition. 0.4mg/mL NLX makes the anoxia-endurance of mice higher than the first group, 0.04mg/mL NLX makes the anoxia-endurance of mice lower than the first group.

    Comparative Study on Mice's Acute Toxicity of Different Components of Radix Bupleuri and Radix Bupleuri Scorzoneraefolii

    Objective To discuss the effect of different composition of Radix Bupleuri and Radix Bupleuri Scorzoneraefolli on mice's acute toxicity. Methods Respectively prepare the water extracted and alcohol extracted components of Radix Bupleuri and Radix Bupleuri Scorzoneraefolli, and compare the acute toxicity of different components in Radix Bupleuri and Radix Bupleuri Scorzoneraefolli on mice in accordance with classical acute toxicity test Methods, and the experimental datas are calculated MTD and MLD with the Kg body weight method, with the continuous administration and observa-tion for 14 days. Results The acute toxicity of different components in Radix Bupleuri and Radix Bupleuri Scorzoneraefolli on mice is: alcohol extracted components of Radix Bupleuri Scorzoneraefolli >alcohol extracted components of Radix Bupleuri >water extracted components of Radix Bupleuri Scorzoneraefolli >water extracted components of Radix Bupleuri. The water extracted components of Radix Bupleuri and Radix Bupleuri Scorzoneraefolli did not appear dead. Calculated by crude drug content, the MLD were respectively 181.6 g·kg-1·d-1 and 201.6 g·kg-1·d-1,which is equal to 1412.4 times and 1568.0 times of 70kg people's daily dried medicinal herb expenses. The alcohol extracted components of Radix Bupleur and Radix Bupleuri Scorzoneraefolli are unable to make LD50, the MTD were respectively 46.4 g·kg-1·d-1 and 75.2 g·kg-1·d-1,which is equal to 360.9 times and 584.9 times of 70kg people's daily dried medicinal herb expenses. Conclusion The toxicity of Radix Bupleuri Scor-zoneraefolli is bigger than the Radix Bupleur, and the toxicity of the alcohol extracted components is bigger than the water extracted components. There is some relevance between the content of Bupleurum saikosaponin and the toxicity, so we speculated that the Bupleurum saikosaponin may be the mainly toxical basis.

    Objective To compare the content of saikosaponin and the acute toxicity on the all components, alcohol-water extracts in different place's Bupleurum, and discuss the effect of original place on the content and toxicity of Bupleurum saikosaponin substances. Methods UV method, HPLC method and the classical method of acute toxicity,were used to determine the content of total saikosaponin and saikosaponin a of the all components, alcohol-water extracts in different places' Bupleurum, and compare the content and the acute toxicity. Results The content of total saikosaponin on the all components in different place's Bupleurum is: Hebei Bupleurum>Inner Mongolia Bupleurum> Shanxi Bupleurum>Henan Bupleurum, and the content of saikosaponin a is: Hebei Bupleurum>Inner Mongolia Bupleurum, Shanxi Bupleurum>Henan Bupleurum, and the MLD is: Henan Bupleurum>Shanxi Bupleurum>Inner Mongolia Bupleurum>Hebei Bupleurum; the content of total saikosaponin on the alcohol-water extracts in different place's Bupleurum is:Hebei Bupleurum>Inner Mongolia Bupleurum>Shanxi Bupleurum>Henan Bupleurum, and the content of saikosaponin a is:Hebei Bupleurum>Shanxi Bupleurum>Inner Mongolia Bupleurum>Henan Bupleurum, and the MTD is:Inner Mongolia Bupleurum>Shanxi Bupleurum>Henan Bupleurum>Hebei Bupleurum. The content of total saikosaponin and saikosaponin a in the all components, alcohol-water extracts of Hebei Bupleurum is higher than other different original place's Bupleurum, and the MLD of the all components and the MTD of the alcohol-water extracts is smaller than other different original place's Bupleurum, but the toxicity is the biggest. Conclusion The original place has some influence on the content of the saikosaponin substances and the toxicity of Bupleurum, and the toxicity of Bupleurum has some relevance with the content of saikosaponin substances.

    Objective To compare the content of saikosaponin and the acute toxicity on Radix Bupleuri Scorzoner-aefolliand its processed products, and discuss the effect of processing on the content and toxicity of Bupleurum saikosaponin substances. Methods HPLC method and the classical method of acute toxicity were used to determine the content of saikosaponin a and the acute toxicity of the Radix Bupleuri Scorzoneraefolliand its processed products, and compare the content and the acute toxicity. Results The content of saikosaponin a on the different processed products which included health products, fried products, vinegar products, wine products and honey roasted products was separately 0.46%,0.16%,0.22%,0.22%,0.32%. The health products, fried products, vinegar products, wine products and honey roasted products of Radix Bupleuri Scorzoneraefolli did not appear dead. Calculated by crude drug content, the MLD were respectively 25.2,28.8,26.4,26.0,32.4g·kg-1,which were respectively equal to 196,224,205,233,252 times of 70kg people's daily dried medicinal herb expenses. Making continuous observation of 14 days, mice were generally in good condition,and no one died. Conclusion The content of saikosaponin a and the acute toxicity of the Radix Bupleuri Scorzoneraefolliand its processed products were different, and no death occurred in each sample,the MLD showed the basic security and low toxicity of the Radix Bupleuri Scorzoneraefolliand its processed products, suggesting that processing has some influence on the content of the saikosaponin substances and the safe dose range of Radix Bupleuri Scorzoneraefolli herbs.

    Study on the Refining Process of the Total Bupleurum Saikosaponin under the Guidance of Toxicity

    Objective By comparing the acute toxicity of mice, to explore the enrichment of different purification process on the total Bupleurum saikosaponin which is the mainly toxical substance basis of Bupleurum. Methods Make the comparing study on mice's acute toxicity of the crude extracts of total Bupleurum saikosaponin, the water elution of total Bupleurum saikosaponin and the alcohol elution of toatal Bupleurum saikosaponin in accordance with classical acute toxicitytest Methods. Results Caculated by total saikosaponin, the LD15 of the crude of total Bu-pleurum saikosaponin is 0.3g·kg-1,and the MTD is 0.48 g·kg-1; the LD10 of the water elution of total Bupleurum saikosaponin is 0.83g·kg-1,and the MTD is 1.324g·kg-1; the LD100 of the alcohol elution of total Bupleurum saikosaponin is 12.4g·kg-1. Conclusion The enrichment of different purification process on the total Bupleurum saikosaponin: the alcohol elution of toatal Bupleurum saikosaponin > the water elution of total Bupleurum saikosaponin > the crude extracts of total Bupleurum saikosaponin;the acute toxicity of different purification process on the total Bupleurum saikosaponin: the alcohol elution of toatal Bupleurum saikosaponin > the crude extracts of total Bupleurum saikosaponin> the water elution of total Bupleurum saikosaponin, so we determine that the best refining process of enriching the total Bupleurum saikosaponin is alcohol elution process.

    Objective By combing pertinent literature,and summarizing the current states of chemical composition of evodiamine related with its efficacy and toxicity, to identify its effective and toxical composition, offer the accordance and research ideas for further research on the relationship between efficacy, toxicity and chemical composition of evo-diamine. Methods The related references in recent 10 years both at home and abroad about evodiamine were analyzed, collated and summarized. Results Alkaloids are the main chemical constituents and active ingredients, and are the main material basis for efficacy in evodiamine; evodiamine has small toxicity documented in pharmacopoeia and lit-erature, but the research reports related to the toxicity of chemical constituents are very few and the research about toxicology of evodiamine is still not deep enough. Conclusion There is or not other effective chemical composition in evodiamine, and what is the toxical chemical composition in evodiamine, and the relevance between effective chemical composition and toxical chemical composition, and the "dose-efficacy" and "dose-toxicity"relationship between efficacy, toxicity and chemical composition in evodiamine, and so on; they are to be further deep study and to provide experimental evidence for the optimization、security and quality control、safety and rational use in clinic.

    Objective To provide the basis for strengthening the special drug administration and establishing counter-measures for the relevant departments of the local government,we learning about the abuse prevalence status of medical narcotic, psychotropic drug and behavioral characteristics of drug abuse in the monitoring reports of Henan Province from 2005 to 2009. Methods According to the Drug Abuse Monitoring Survey Form which was editeds by the national drug abuse monitoring centers, we monitor drug abusers who were received to drug treatment agencies from 2005 to 2009, in which medical used narcotic, psychotropic drug abuse will be the major survey. Results The categories of the abused medical narcotic, psychotropic drug show a rising trend, and the kind of abused medical narcotic and psychotropic substances has rised from 10 in 2005 to 15 in 2009. Conclusion The main abused medical of narcotic, psychotropic substances include meperidine, stability, morphine, triazolam, tramadol, while other medical narcotic, psychotropic substances and some prescription drugs also exist.

    Objective To study the perioperative use of prophylactic antibiotics in the hospita1. Methods 355 surgery patients were sampled randomly from Jun.to Dec.2008 in the hospital and investigated in respect of the perioperative use of prophylactic antibiotics based on the Guidir,g Principle of the CHnical Application of Antibacterials Results The general surgical department of antibacterials took up 100%, antibiotics involved in 9 Classes 32 varieties, of which 327 cases of preventive medicine, accounting for 92.11%, treatment drugs in 28 cases, accounting for 7.89%. All analytical cases, a single medication, two united agents, triple agents, four joint and four or more medications together accounted for 24.51%, 46.48%, 17.46%, 11.55%. Conclusion We should strengthen the use of perioperative anti-infective management and supervision, promote rational drug clinicians.

    According to the recent Results of clinical trials and literatures, some problems were discussed in this paper, such as the co-administrasion of cardiovascular drugs, drug adverse reactions and the rational use of medicines. Meanwhile, we discussed the rational time and dosage that patients should pay attention to,especially for some special patients, like female patients and old age patients whose physiologic function has some difference with others. Some questions about how to choose the best cardiovascular drugs and dosage were discussed also. The aim is to improve the rationality of clinical medication and reduce the ADR risk.

    Analysis of 104 Cases of Adverse Drug Reaction Reports of Antibacterial Agents

    Objective To investigate the situation of adverse drug reaction/event(ADR/ADE) of antibacterial agents, thus to provide bases and references for rational use of antibacterial agents. Methods 104 ADR reports of antibacterial agentswere collected and analyzed in a hospital during 2009. Result Of the total 104 ADR reports, females was slightly more than males; adult patients between 18 to 59 years old age were accounted for the majority; 72 cases that administered intravenously were accounted for the majority(69.23%); the cases that caused by cephalosporins were accounted for the most(40 cases,38.46%), and damage to the skin and accessories(54 cases, 51.92%)was the commonest type. Conclusion We should pay great attention to the ADR of antibacterial agents, and strengthen the supervision of clinical application of antibacterial agents.

    Analysis on 161 New or Serious Reports of Xixinnao Injection

    Objective of the past 4 years Xixinnao injection adverse reaction/event(ADR/ADE) reports were reviewed, provide a reference for clinical medication. Methods Jinan ADR Monitoring Center received 161 cases 2006~2009 Xixinnao injection of new, serious(the new strict) of ADR/ADE reports were collected and analyzed. Results Xixinnao injection of 161 patients with new stringent ADR/ADE cases occurred within 20min of time in 129 drug cases, accounting for 80.12%; children as high risk group, accounting for 49.69% in all cases. Xixinnao injection production process of accessories, the stability of the drug itself, and the combination of clinical medicine such as the joint leading to ADR/ADE of the main reasons. Conclusion Xixinnao injection in clinical ADR/ADE should attract great attention of clinical medicine; proposed system for pharmacy instructions. Hope that the security departments of the drugs should be further studied.
