Study on Inhibition Effect and Mechanism of Baicalin on Human Gastric Carcinoma SGC-7901 Cells

Abstract: Objective To investigate the inhibiting effect and its mechanism of baicalin on human gastric carcinoma SGC-7901 cells. Methods Growth inhibition rates of SGC-7901 cells were measured with MTT method, the cell cycle distribution was detected by PI single staining, apoptosis was detected by Annexin V/PI double staining, and the expression levels of Bcl-2, bax protein were analyzed by western blot. Results Baicalin inhibited the proliferation of SGC-7901 cells ina dose and time dependent manner. The IC50 of 24h and 48 h were 218.73 μmol·L-1 and 161.86μmol·L-1 respectively. The cell growth was arrested in G2/M phase. Fluorescent staining can detect the apoptotic cells induced by baicalin. The expression of Bax protein was increased while the Bcl-2 was decreased. Conclusion Baicalin not only inhibits proliferation but also induces apoptosis of SGC -7901 cells. Its mechanism is related to down -regulating Bcl-2 while up-regulating Bax.
