Effects of Matrine on Proliferation and Metastasis of Human Endometrial Carcinoma Cell Line Ishikawa

Abstract: Objective To study the effect of Matrine on inflammatory factor IL-8 and NF-кBp65 expression from human endometrial carcinoma cell line Ishikawa. Methods The changes of cell proliferation of Ishikawa cells treated with Matrine were observed by using MTT assay. The cell cycle expressions were determined by flow cytometry.The densities of IL-8 in the supenmatant fluid were measured by ELISA. And NF-кBp65 were determined by western blot assay. Results Matrine could inhibit the proliferation of Ishikawa cells in vitro significantly in a dose and time dependent manner(P<0.05). The G0/G1 phase arrest was induced by treatment after 48 hours(P<0.05). The densities of IL-8 in the supenmatant fluid were decreased. Results revealed that cellular NF-кBp65 levels decreased in Matrine-treated cells at 48 hours(P<0.05). Conclusion Matrine could inhibit Ishikawa cell proliferation, induce G0/G1 phase arrest and by interfering with the NF-кB signaling pathway.
