汉防己甲素对MCF-7 细胞Akt激酶活性影响的研究

Study on Effect of Tetrandrine on Akt Activity in MCF-7 Cell Line

摘要: 目的 探讨汉防己甲素诱导人乳腺癌细胞MCF-7凋亡的分子机制。方法 采用流式细胞法测定不同浓度汉防己甲素(1、5、10 μmol·L-1)诱导MCF-7凋亡率;免疫印迹法检测汉防己甲素对MCF-7细胞总Akt及p-Akt ser437、p-Akt Thr308表达的影响;Akt激酶活性检测试剂盒检测Akt激酶活性的改变。结果 不同浓度的Tet(1、5、10 μmol·L-1)对培养的乳腺癌细胞MCF-7可有效的诱导凋亡;同时Tet可以有效的抑制p-Akt ser437、p-Akt Thr308蛋白表达,但对总Akt蛋白的表达没有影响。Akt 激酶活性的分析显示:不同浓度的Tet处理MCF-7细胞48 h后,磷酸化的GSK3α/β蛋白浓度逐渐下降,间接提示Akt激酶活性下降。结论 汉防己碱可能通过抑制Akt激酶活性并影响其下游的信号途径诱导乳腺癌细胞凋亡。

Abstract: Objective To study the cell signal mechanism of tetrandrine-induced apoptosis of human breast cancer cell line MCF-7. Methods The effects of tetrandrine-induced apoptosis of MCF-7 (1, 5, 10 μmol·L-1) was detected by flow cytometry. Western blot was performed to assess expression of Akt, p-Akt ser437 and p-Akt Thr308 in MCF-7. The activity of Akt was detected by Akt activity assay kit (KinaseSTAR). Results Different doses of tetrandrine (Tet) could induce apoptosis of MCF-7 in a dose-dependent manor. Tet could significantly inhibit expression of p-Akt ser437and p-Akt Thr308, but had no effect to expression of total Akt in MCF-7. Tet could down-regulate the activity of Akt of MCF-7. Conclusion Tet can induce apoptosis of MCF-7 cells through suppressing Akt activity.
