Abstract: Objective To explore the influence of Xiaoerganmaoning granules (XRG) on the express of MCP-1, ERK and NF-κB signaling pathway in influenza virus infected mice. Methods The mice were infected with influenza virus FM1 and PR8 strain. The treatment groups were given 3 doses levels of Xiaoerganmaoning Granules (28, 14 and 7 g·kg-1). The ribavirin was the positive drug control. After 5 days, enzyme-linked immunoassay (ELISA) method was adopted to study the content of MCP-1 in serum of influenza virus PR8 infected mice. The western blot was used to observe the protein expression of ERK and NF-κB signaling pathway in lung tissue of influenza virus FM1 infected mice. Results Compared with the model control group, the content of MCP-1 was decreased significantly by XRG. and the content of MCP-1 in the 3 XRG groups (P<0.05). Compared with the model control group, the protein expression of p-Erk in XRG medium dose group was observably reduced (P<0.05). The 3 XRG groups had the tendency of reducing the expression of NF-κB.Conclusion XRG could alleviate the inflammatory reaction and exert antiviral effect by inhibiting the MCP-1, ERK and NF-κB signaling pathway to enhance the immune function and control the proliferation of influenza virus in vivo.
