抗病毒口服液治疗甲型H1N1流感病毒感染药效及机制探讨

Treatment Effects and Mechanisms of Kangbingdu Oral Liquid Against the Infection of Influenza A (H1N1)

摘要: 目的 观察抗病毒口服液对体内感染甲型H1N1流感病毒模型的治疗作用并探讨其作用机制。方法 使用ICR小鼠,采用甲型H1N1流感病毒FM1株及PR8株经滴鼻感染小鼠造成肺炎模型,经治疗给药后,计算各组小鼠肺指数;采用实时荧光定量聚合酶链式反应RT-PCR法检测小鼠肺组织流感病毒载量;采用ELISA法检测小鼠血清中丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)和γ干扰素(IFN-γ)的含量;体外采用甲型H1N1流感病毒FM1株及PR8株感染MDCK细胞,经治疗给药后,检测细胞上清液中神经氨酸酶(NA)活性。结果 与模型组相比,抗病毒口服液在体内能明显降低流感病毒感染小鼠的肺指数,降低肺组织中的流感病毒载量,降低TNF-α的含量并升高IFN-γ的含量;在体外可明显抑制NA活性,且均具有统计学意义(P <0.05或P <0.01)。结论 抗病毒口服液对甲型H1N1流感病毒体内感染有较好的治疗作用,其作用机制可能在于抑制炎性反应并抑制病毒繁殖。

Abstract: Objective To observe the treatment effects and mechanisms of Kangbingdu oral liquid against influenza A (H1N1) virus infection. Methods ICR mice were infected by influenza A (H1N1) virus FM1 strain and PR8 strain. After treatment, the lung index, virus load and the level of MDA、TNF-α and IFN-γ in serum were measured; MDCK cells were infected by influenza A (H1N1) virus, after treatment, the activation of neuraminidase(NA) was measured. Results Compared with model group, Kangbingdu oral liquid significantly reduced the lung index of mice, significantly reduced the virus load, inhibited the level of TNF-α and increased the level of IFN-γ in vivo; Kangbingdu oral liquid significantly inhibited the activation of NA in vitro (P<0.05 or P<0.01). Conclusion Kangbingdu oral liquid has good therapeutic effects on viral pneumonia caused by influenza A (H1N1) virus through the inhibition of inflammatory response and virus copy.
