吴茱萸水提组分镇痛及伴随毒副作用机制研究

摘要: 目的 对吴茱萸水提组分的镇痛机制进行初步研究,探讨其伴随毒性与氧化损伤机制的相关性。方法 小鼠按体重随机分为5 组,即正常对照组、扶他林组、吴茱萸水提组分高、中、低剂量组,连续给药7 天,末次药后90min 取血检测前列腺素E2(PGE2)水平,检测血内丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、一氧化氮(NO)、一氧化氮合酶(NOS)、谷胱甘肽(GSH)和谷胱甘肽过氧化物酶(GSH-Px)的含量和活性。结果 吴茱萸水提组分可致血中PGE2 含量水平明显降低,可致血中MDA 含量增加,同时SOD 活性下降;血中一氧化氮(NO)含量增加,一氧化氮合酶(NOS)活性升高;血中谷胱甘肽(GSH)含量下降,谷胱甘肽过氧化物酶(GSH-Px)活性下降。上述变化趋势随剂量增加而加重,呈现一定的剂量依赖关系,与空白对照组比较有明显差异。结论 吴茱萸水提组分镇痛机制主要与降低血中PGE2 含量有关,其伴随肝毒性损伤途径与引起机体氧化应激后诱导脂质过氧化有关。

Abstract: Objective To make the preliminary study on analgetic effect mechanism of water extraction components of Evodia Fructus, and to discuss the relevance between the accompanying hepatotoxicity and oxidative damage mechanism. Methods Mice were randomly divided into 5 groups according to weight, namely normal group, diclofanac sodium sustained release tablets group, high, middle and low dose group of water extraction components of Evodia Fructus. Mice were continuously administrated for7 days, and the level of PGE2, MDA、SOD、NO、NOS、GSH、 GSH-Px in serum were detected. Results The water extraction components of Evodia Fructus could cause the level of PGE2 in serum decreased, the activity of MDA in serum increased, and the activity of SOD decreased. The level of NO and NOS were increased, and the activity of GSH and GSH-Px were decreased. All the changes were aggravated in accordance with the dosages, and there was sijnificant difference compared with distilled water control group and there was certain dose -dependence relationship. Conclusion The analgetic effect mechanism of water extraction components of Evodia Fructus was mainly related with the reduction of the content of PGE2 in serum, and the approach of accompanying hepatotoxicity damage was related with the peroxidative damage mechanism.
