HPLC法同时测定刺五加果实中齐墩果酸和熊果酸的含量

Simultaneously Determination of Oleanolic Acid and Ursolic Acid in Medofenoxate Fruits by HPLC

摘要: 目的 建立HPLC 同时测定刺五加果实中齐墩果酸和熊果酸含量的方法。方法 采用Agilent XDB-C18色谱柱(4.6mm×250mm,5 m),以甲醇- 乙腈-0.3%磷酸溶液(30 60 10)为流动相,流速:0.8mL·min-1,柱温为室温,检测波长为210nm。结果 齐墩果酸进样量在0.2052~3.2832 g范围内线性关系良好,r=0.9995,平均加样回收率为101.11%,RSD 为1.69%(n=6);熊果酸进样量在0.2072~3.3152 g范围内线性关系良好,r=0.9993,平均加样回收率为98.51%,RSD 为2.67%(n=6)。结论 该方法灵敏快速、结果准确、重现性好,可用于刺五加果实中齐墩果酸和熊果酸含量的同时测定。

Abstract: Objective To establish a HPLC method for the simultaneous determination of Oleanolic Acid and Ursolic Acid in Medofenoxate fruits. Methods Agilent XDBC18(4.6 mm×250 mm,5μm) column was applied, using methanolacetonitrile-0.3% phosphoric acid(30 :60 :10) as mobile phase with the rate of 0.8mL·min-1. The column temperature was room temperature and the detective wavelength was 210nm. Results A good linearity was obtained over the range of 0.2052~3.2832μg for Oleanolic Acid(r=0.9995). The average recovery(n=6) was 101.11%(RSD=1.69%);A good linearity was obtained over the range of 0.2072~3.3152μg for Ursolic Acid(r=0.9993). The average recovery(n=6) was 98.51%(RSD=2.67%). Conclusion The method is practical, convenient, accurate, and can be used as the simultaneous determination of Oleanolic Acid and Ursolic Acid in Medofenoxate fruits.
