一测多评法用于橘核中多成分含量测定的适用性研究

摘要: 目的 建立一测多评法同时测定橘核中柠檬苦素、诺米林、黄柏酮3种成分含量的方法,并验证该法在橘核质量控制中应用的可行性与适用性。方法 橘核含量测定采用高效液相色谱法,以Waters Symmetry C18柱(4.6×250 mm, 5 μm)为色谱柱,流动相为乙腈-水(45∶55),流速为1 mL/min,柱温35 ℃,检测波长为220 nm。以柠檬苦素为内参物,计算诺米林、黄柏酮的相对校正因子,测定橘核中3种成分的含有量。结果 建立了橘核中柠檬苦素与诺米林、黄柏酮的相对校正因子;3种成分一测多评法的计算值与外标法实测值间无明显差异。结论 一测多评的质量控制模式可用于橘核中3种不同成分的多指标同步质量评价。

Abstract: Objective To establish a quantitative analysis of multi-components by single mark (QAMS) for determining the contents of three compositions (limonin, nomilin, obacunone) from orange kernels, salt citri reticulatae semen and their granule, and verify the feasibility and applicability of this method in quality control. Methods Limonin was used as the internal reference substance. The HPLC analysis was performed on a Waters Symmetry C18 (4.6×250 mm, 5 μm) column with a mobile phase consisted of acetonitrile and water (45∶55) at a flow rate of 1 mL/min-1. The column temperature was maintained at 35 ℃, and the detection wavelength was set at 220 nm. Results The relative correction factors of Limonin, nomilin and obacunone were established, without significant difference between the calculated values by QAMS and measured valves. Conclusion The experimental QAMS method can accurately determine the contents of the three compositions in citri reticulatae semen, and can be used in the multi-index evaluation in orange kernels.
