平贝母和给药平贝母后大鼠尿液内化学成分LC-MSn分析

LC-MSn Analysis of the Components in Bulbus Fritillariae Ussuriensis and the Urine of Rats after Oral Administered Bulbus Fritillariae Ussuriensis

Abstract: Objective To investigate the components in Bulbus Fritillariae ussuriensis by LC-MSn, and to quickly identify components in rats urine after orally administered Bulbus Fritillariae ussuriensis. Methods The structures of components in Bulbus Fritillariae ussuriensis were detected by LC-MSn experiments, combined with the same kind of compound standard of mass spectrometry cracking rule, identification of each chromatographic peak, then identify the urine within the same way. Results 11 components were identified from Bulbus Fritillariae ussuriensis, and 8 components were identified from the urine of rats after orally administered Bulbus Fritillariae ussuriensis. Conclusion This method can be used for accurate identification of steroidal alkaloid components in Bulbus Fritillariae ussuriensis and the urine of rats after orally administered Bulbus Fritillariae ussuriensis.

摘要: 目的建立平贝母化学成分的液质联用分析方法,并以此快速指认大鼠给药平贝母后尿液内化学成分。方法运用LC-MSn联用技术得到贝母水煎液内各化合物的总离子流图和多级质谱图,结合同种类化合物标准品质谱裂解规律,从而对各色谱峰进行鉴定,并以此指认尿液内药源成分。结果从平贝母水煎液中鉴定出11个化合物,从大鼠给药平贝母尿液中鉴定出8种化合物。结论HPLC-MSn法可用于平贝母中异甾体类生物碱的结构分析,并可用于快速推断给药平贝母后尿液内异甾体类生物碱成分,为建立准确的质量评价方法和生物样品快速指认提供了参考。
