超声波优化栀子苷提取的工艺研究

Optimization of Ultrasonic Extraction Conditions of Geniposide

摘要: 目的 研究栀子苷的最佳超声提取条件。方法 采用响应面分析法优化栀子苷的超声提取条件,在单因素实验基础上利用BBD组合设计Box-Behnken 中心组合设计拟合试验条件。结果 以栀子苷提取率为响应值进行多元二次响应回归分析,可得超声提取栀子苷的最佳工艺条件为:超声功率 750.87 W、提取时间 46.57 min、溶剂浓度 52.06 %、溶剂体积(料液比)1:64.09 g·mL-1。此最优条件下栀子苷提取率预测为 36.16 mg·g-1,验证值为 35.49 mg·g-1,与预测值的相对误差为 1.85%。结论 此最优工艺条件具有一定的实际应用价值,对工业化生产有一定的指导意义。

Abstract: Objective To study the technology of extracting geniposide from Gardenia jasminoides Ellis by ultrasound extraction. Methods Based on the single-factor experiments, a four-factor-three-level experiment design was developed by Box-Behnken central composite design method. Multi-quadratic equation was established with the yield of geniposide as response values. Results The optimum extraction condition was obtained as follows: ultrasound power 750.87 W, extraction time 46.57 min, ethanol concentration 52.06 % and ethanol volume(ratio of solvent to solid) 1:64.09 g·mL-1. The predictive maximum extraction yield was 36.16 mg·g-1 while the average experimental value was 35.49 mg·g-1, with the relative error of 1.85%. Conclusion The method is reliable, and can be used for industrial production.
