Protective Effect of Yindan Xinnaotong Capsule on Endothelial Cells in Vitro

Abstract: ObjectiveTo study the protective effect of Yindan Xinnaotong capsule(YXC) on endothelial cells. MethodsHuman umbilical vein endothelial cells (HUVECs) were cultured in vitro and HUVECs were pretreated with 250 μg·mL-1,125 μg·mL-1 and 62.5 μg·mL-1 for 24 h. 200 μg·mL-1 Ox-LDL and 60 ng·mL-1 TNF-α were used to induce HUVECs injury. The cell survival rate was determined by MTT, and flow cytometry was used to detect apoptosis to evaluate the mechanism of protective function of YXC. ResultsThe maximal non-cytotoxic concentration of YXC was 500 μg·mL-1, thus 250, 125 and 62.5 μg·mL-1 were employed as the experimental concentrations. YXC could obviously increase the survival rate of HUVECs induced by Ox-LDL (P <0.05) and TNF-α with a dose-dependence manner; compared with model group, 62.5 and 250 μg·mL-1 YXC treatment decreased apoptosis rate induced by Ox-LDL HUVECs (P <0.05). Meanwhile, 250 and 125 μg·mL-1 YXC obviously reduced apoptosis rate induced by TNF-α (P <0.05). ConclusionYXC could protect the HUVEC injury induced by Ox-LDL and TNF-α, and this may be associated with the inhibitory effect on cell apoptosis.
