石榴皮鞣质对膀胱癌T24细胞迁移侵袭能力的影响研究

Effect of Pomegranate Peel Tannin on Migration and Invasion of Bladder Cancer T24 Cells

摘要: 目的 探讨石榴皮鞣质对人膀胱癌T24细胞迁移及侵袭能力的影响。方法 以人膀胱癌T24细胞为靶细胞,通过细胞划痕试验及Transwell迁移试验观察不同浓度石榴皮鞣质对T24细胞迁移能力的影响;采用Transwell侵袭试验测定石榴皮鞣质对T24细胞侵袭能力的影响。结果 细胞划痕试验表明,石榴皮鞣质作用后T24细胞伤口愈合能力显著下降,划痕后48 h空白对照组的伤口愈合率为52.04 %,12.5μg ·mL-1 石榴皮鞣质组的愈合率为18.21 % ,与空白对照组相比差异具有统计学意义(P < 0.001)。Transwell迁移试验显示,经石榴皮鞣质处理24 h 后T24细胞穿膜细胞数明显减少并呈浓度依赖性(P < 0.001)。Transwell侵袭试验结果证明石榴皮鞣质组穿过人工基底膜的细胞数显著减少,可降低侵袭活性(P < 0.001)。结论 石榴皮鞣质具有较好抑制膀胱癌细胞转移的活性。

Abstract: Objective To investigate the effect of pomegranate peel tannin on the migration and invasion of human bladder cancer T24 cells. Methods Human bladder cancer T24 cells were used as target cells, and the effects of different concentrations of tannin on migration of T24 cells were observed by cell scratch test and Transwell migration test. Transwell invasion assay was used to determine the effect of pomegranate peel tannin on the invasion ability of T24 cells. Results The results of cell scratch test showed that the wound healing ability of T24 cells decreased significantly after pomegranate peel tannin treatment. After 48 h, the wound healing rate of blank control group was 52.04 %, 12.5 μg·mL-1 pomegranate peel tannin group was 18.21 %, and the differences were statistically significant compared with the blank control group (P <0.001). Transwell migration assay indicated that the number of T24 cells passing through the membrane was significantly decreased after treated with pomegranate peel tannin for 24 h (P <0.001). The results of Transwell invasion test showed that the number of cells in the pomegranate peel tannin group passing through the artificial basement membrane decreased significantly. It can decrease the invasive activity (P <0.001). Conclusion Pomegranate peel tannin can inhibit the metastasis of bladder cancer cells.
