Study on Mechanism of Cytokine and NO Injury in the Hepatotoxical Injury in Rats Caused by Volatile Oil from Bupleurum Chinense

摘要: 目的 对细胞因子及NO在柴胡挥发油致大鼠肝损伤中的作用机制进行研究,为阐明柴胡挥发油的肝损伤机制提供实验依据。方法 连续15天给大鼠灌胃不同剂量的柴胡挥发油,末次药后分别检测大鼠血清和肝组织iNOS活性以及NO、TNF-α、IL-6 、IL-10含量的变化。结果 柴胡挥发油可致大鼠血清和肝组织iNOS活性升高,NO、TNF-α、IL-6 、IL-10含量均增加,与正常对照组比较有不同程度的显著性差异。结论 TNF-α、IL-6 、IL-10等细胞因子可介导肝细胞的损伤,NO可加重肝毒性损伤。

Abstract: Objective To study the effects of cytokine and NO on the hepatotoxical injury caused by volatile oil from Bupleurum Chinense and offer experimental basement for the explanation of mechanisms of hepatotoxical injury. Methods Volatile oil from Bupleurum Chinense of different dosages was given to Wistar rats by intragastric administration for fifteen days consecutively. The activity of iNOS and contents of TNF-α,IL-6,IL-10 and NO both in serum and liver were tested after the last administration. Results The siginificant increase of activity of iNOS and contents of TNF-α,IL-6,IL-10 and NO both in serum and liver can be induced by volatile oil from Bupleurum Chinense. Conclusion Hepatotoxical injury caused by volatile oil from Bupleurum Chinense can be mediated via cytokine. Nitric can aggravate the injury but the path by which it reacted is left to be studied.
