HPLC法测定东北铁线莲中白头翁素的含量

Determination of the Contents of Anemonin in Clematis mandshurica Rupr by HPLC

摘要: 目的 建立东北铁线莲中白头翁素高效液相色谱法的测定方法。方法 采用Agilent XDB-C18色谱柱(250mm×4.6mm,5μm),以甲醇-水(40:60)为流动相,流速:0.8mL·min-1,柱温为室温,检测波长为220nm。结果 白头翁素在5.45~109μg·mL-1范围内线性关系良好,r=0.9993,平均加样回收率为102.41%,RSD为2.78%(n=6)。结论 该方法操作简单,灵敏,重现性好,可以用来测定东北铁线莲中白头翁素的含量。

Abstract: Objective To establish a HPLC method for the determination of anemonin in Clematis mandshurica Rupr. Methods Agilent XDB C18(4.6 mm×250 mm, 5μm) column was applied,using. methanol-water(40:60) as mobile phase with the rate of 0.8mL·min-1. The column temperature was room temperature and the detective wavelength was 220 nm. Results A good linearity was obtained over the range of 5.45~109μg·mL-1 for anemonin (r=0.9993). The average recovery(n=6) was 102.41%(RSD=2.78%). Conclusion The method is simple, sensitive,good reproducibility and used as the determination of Anemonin in Clematis mandshurica Rupr.
