毒性导向下的柴胡总皂苷精制工艺研究

Study on the Refining Process of the Total Bupleurum Saikosaponin under the Guidance of Toxicity

摘要: 目的 通过小鼠急性毒性比较研究,探讨不同精制工艺对毒性物质基础柴胡总皂苷的富集程度。方法 参照经典的急性毒性实验方法,进行柴胡总皂苷粗提取物、柴胡总皂苷水洗脱液和柴胡总皂苷醇洗脱液对小鼠的急性毒性比较研究。结果 按总皂苷含量计算,测得柴胡总皂苷粗提取物LD15为0.3 g·kg-1,MTD为 0.48 g·kg-1;测得柴胡总皂苷水洗脱液LD10为0.83 g·kg-1,MTD为 1.324 g·kg-1;测得柴胡总皂苷醇洗脱液LD100为12.4 g·kg-1结论 上述不同精制工艺样品的柴胡总皂苷富集程度为:柴胡总皂苷醇洗脱液 > 柴胡总皂苷水洗脱液 > 柴胡总皂苷粗提物 ;上述不同精制工艺样品的小鼠急性毒性大小为:柴胡总皂苷醇洗脱液 > 柴胡总皂苷粗提物 > 柴胡总皂苷水洗脱液,故确定醇洗脱工艺为富集柴胡总皂苷的最佳精制工艺。

Abstract: Objective By comparing the acute toxicity of mice, to explore the enrichment of different purification process on the total Bupleurum saikosaponin which is the mainly toxical substance basis of Bupleurum. Methods Make the comparing study on mice's acute toxicity of the crude extracts of total Bupleurum saikosaponin, the water elution of total Bupleurum saikosaponin and the alcohol elution of toatal Bupleurum saikosaponin in accordance with classical acute toxicitytest Methods. Results Caculated by total saikosaponin, the LD15 of the crude of total Bu-pleurum saikosaponin is 0.3g·kg-1,and the MTD is 0.48 g·kg-1; the LD10 of the water elution of total Bupleurum saikosaponin is 0.83g·kg-1,and the MTD is 1.324g·kg-1; the LD100 of the alcohol elution of total Bupleurum saikosaponin is 12.4g·kg-1. Conclusion The enrichment of different purification process on the total Bupleurum saikosaponin: the alcohol elution of toatal Bupleurum saikosaponin > the water elution of total Bupleurum saikosaponin > the crude extracts of total Bupleurum saikosaponin;the acute toxicity of different purification process on the total Bupleurum saikosaponin: the alcohol elution of toatal Bupleurum saikosaponin > the crude extracts of total Bupleurum saikosaponin> the water elution of total Bupleurum saikosaponin, so we determine that the best refining process of enriching the total Bupleurum saikosaponin is alcohol elution process.
